Summary Factors exhibiting anti-complementary activity released from trypanosomes after incubation at 20°C were described. The active material was shown to consume the first component of bovine complement. While the anticomplementary factor(s) from T. lewisi could activate bovine, human and guinea pig complement, the factor(s) from T. congolense was observed to activate bovine complement, but not guinea pig and only slightly human complement. The roles of complement activating factor(s) of trypanosomes in the pathology of the disease are discussed.This project is supported by National Research Council of Canada grant A 0068 and a grant from the International Development Research Centre.  相似文献

Protein amyloid is often deposited in connection with neurodegenerative diseases. Such deposits generally possess three principal drawbacks: cytotoxicity, lack of spatial control in their deposition and structural polymorphism. These are typical features of biologically non-optimized systems which have not been exposed to evolutionary pressure. Nevertheless, Nature uses the cross-beta self-organizing principle in many structural contexts where a strong but pliable material is needed. Functional amyloid is found in humans, invertebrates, fungi and, not least, bacteria, in which amyloid may be the rule rather than the exception. Detailed case studies reveal how directed nucleation can use tailor-made proteins optimized to assume a specific amyloid conformation, leading to remarkably robust assemblies. This makes it highly challenging to purify and analyze the products formed in vivo. We contrast pathogenic and in-vitro-formed amyloid with functional amyloid, paying particular reference to bacterial amyloid, and discuss challenges and perspectives in identifying and characterizing this class of protein.  相似文献

As population structure can result in spurious associations, it has constrained the use of association studies in human and plant genetics. Association mapping, however, holds great promise if true signals of functional association can be separated from the vast number of false signals generated by population structure. We have developed a unified mixed-model approach to account for multiple levels of relatedness simultaneously as detected by random genetic markers. We applied this new approach to two samples: a family-based sample of 14 human families, for quantitative gene expression dissection, and a sample of 277 diverse maize inbred lines with complex familial relationships and population structure, for quantitative trait dissection. Our method demonstrates improved control of both type I and type II error rates over other methods. As this new method crosses the boundary between family-based and structured association samples, it provides a powerful complement to currently available methods for association mapping.  相似文献

Ocean island basalts are generally thought to be the surface expression of mantle plumes, but the nature of the components in the source regions of such mantle plumes is a subject of long-standing debate. The lavas erupted at Hawaii have attracted particular attention, as it has been proposed that coupled 186Os and 187Os anomalies reflect interaction with the Earth's metallic core. It has recently been suggested, however, that such variations could also result from addition of oceanic ferromanganese sediments to the mantle source of these lavas. Here we show that Hawaiian picrites with osmium isotope anomalies also exhibit pronounced thallium isotope variations, which are coupled with caesium/thallium ratios that extend to values much lower than commonly observed for mantle-derived rocks. This correlation cannot be created by admixing of core material, and is best explained by the addition of ferromanganese sediments into the Hawaii mantle source region. However, the lack of correlation between thallium and osmium isotopes and the high thallium/osmium ratios of ferromanganese sediments preclude a sedimentary origin for the osmium isotope anomalies, and leaves core-mantle interaction as a viable explanation for the osmium isotope variations of the Hawaiian picrites.  相似文献

Investigators since Freud have appreciated that memories of the people, places, activities and emotions of daily life are reflected in dreams but are typically so fragmented that their predictability is nil. The mechanisms that translate such memories into dream images remain largely unknown. Mass is the most fundamental parameter of a star, yet it is also one of the most difficult to measure directly. In general, astronomers estimate stellar masses by determining the luminosity and using the 'mass-luminosity' relationship, but this relationship has never been accurately calibrated for young, low-mass stars and brown dwarfs. Masses for these low-mass objects are therefore constrained only by theoretical models. A new high-contrast adaptive optics camera enabled the discovery of a young (50 million years) companion only 0.156 arcseconds (2.3 au) from the more luminous (> 120 times brighter) star AB Doradus A. Here we report a dynamical determination of the mass of the newly resolved low-mass companion AB Dor C, whose mass is 0.090 +/- 0.005 solar masses. Given its measured 1-2-micrometre luminosity, we have found that the standard mass-luminosity relations overestimate the near-infrared luminosity of such objects by about a factor of approximately 2.5 at young ages. The young, cool objects hitherto thought to be substellar in mass are therefore about twice as massive, which means that the frequency of brown dwarfs and planetary mass objects in young stellar clusters has been overestimated.  相似文献

During Late Cretaceous and Cenozoic times, many Palaeozoic and Mesozoic rifts and basin structures in the interior of the European continent underwent several phases of inversion (the process of shortening a previously extensional basin). The main phases occurred during the Late Cretaceous and Middle Palaeocene, and have been previously explained by pulses of compression, mainly from the Alpine orogen. Here we show that the main phases differed both in structural style and cause. The Cretaceous phase was characterized by narrow uplift zones, reverse activation of faults, crustal shortening, and the formation of asymmetric marginal troughs. In contrast, the Middle Palaeocene phase was characterized by dome-like uplift of a wider area with only mild fault movements, and formation of more distal and shallow marginal troughs. A simple flexural model explains how domal, secondary inversion follows inevitably from primary, convergence-related inversion on relaxation of the in-plane tectonic stress. The onset of relaxation inversions was plate-wide and simultaneous, and may have been triggered by stress changes caused by elevation of the North Atlantic lithosphere by the Iceland plume or the drop in the north-south convergence rate between Africa and Europe.  相似文献

Positive selection at the molecular level is usually indicated by an increase in the ratio of non-synonymous to synonymous substitutions (dN/dS) in comparative data. However, Plotkin et al. describe a new method for detecting positive selection based on a single nucleotide sequence. We show here that this method is particularly sensitive to assumptions regarding the underlying mutational processes and does not provide a reliable way to identify positive selection.  相似文献
